Parkinson's disease (帕金森氏症) (PD) is the second most common neurodegenerative disease (【醫】神經退化性疾病), affecting over a million people in the United States alone, and is characterized by rigidity (僵硬), bradykinesia (動作遲緩), resting tremor (靜止性的顫抖), and postural instability (姿勢不穩定). Its main neuropathological (神經病理學的) feature is the loss of dopaminergic neurons (多巴胺神經元) of the substantia nigra pars compacta. However, the pathogenesis (致病的原因) of this loss is not understood fully. One of the earliest biochemical changes seen in PD is a reduction in the levels of total glutathione (穀胱甘肽), a key cellular antioxidant. Traditionally, it has been thought that this decrease in GSH levels is the consequence of increased oxidative stress, a process heavily implicated in PD pathogenesis (致病的原因). However, emerging evidence suggests that GSH depletion may itself play an active role in PD pathogenesis (致病的原因). This review aims to explore the contribution of GSH depletion to PD pathogenesis (致病的原因).—Martin, H.
